- Identity
- Macroscopic and microscopic examination, HPTLC fingerprint or DNA barcoding to confirm Grifola frondosa and the part used (fruiting body or mycelium).
- Beta-glucan and alpha-glucan (enzymatic)
- Beta-glucan measured by an enzymatic kit method (such as the Megazyme mushroom and yeast beta-glucan assay), with alpha-glucan reported alongside to show starch from grain substrate. A total polysaccharide figure by UV-Vis does not separate the two.
- Heavy metals
- Lead, arsenic, cadmium and mercury by ICP-MS. Mushrooms take up metals from their growing substrate, so each lot is checked against the buyer's limits.
- Microbiology
- Total plate count, yeast and mold, coliforms, E. coli and Salmonella.
- Pesticide residues
- Multi-residue screen by GC-MS/MS and LC-MS/MS, required for organic material and commonly asked for on conventional lots.
- Physical tests
- Loss on drying, particle size or mesh, and bulk density; for extracts, the extract ratio, carrier and residual ethanol if a solvent was used.
