- Identity
- HPLC or HPTLC anthocyanin profile, where cyanidin-3-sambubioside and cyanidin-3-glucoside dominate in Sambucus nigra. The profile also flags substitution with other dark fruits or added colors.
- Anthocyanin assay
- Total anthocyanins by HPLC or by pH-differential spectrophotometry, sometimes with total polyphenols. The two methods give different numbers, so the spec should name the method.
- Brix, pH and acidity
- For juice concentrate: soluble solids (Brix), pH and titratable acidity, which set dilution and formulation.
- Cyanogenic glycosides
- Sambunigrin or total cyanogenic glycosides, requested by some buyers and markets to confirm adequate processing.
- Heavy metals
- Lead, arsenic, cadmium and mercury, usually by ICP-MS.
- Microbiology
- Total plate count, yeast and mold, E. coli and Salmonella; yeast and mold matter most for concentrates and syrups.
- Pesticide residues
- Multi-residue screen, required for organic claims.
